Target Information
Factor XII is a single-chain zymogen that becomes a two-chain serine protease upon activation, forming heavy (alpha-factor XIIa) and light chains.
The heavy chain contains fibronectin-type, EGF-like, kringle, and proline-rich domains, while the light chain holds the catalytic domain.
Activation leads to further cleavage producing beta-factor XIIa.
Factor XIIa participates in blood coagulation, fibrinolysis, and generation of bradykinin and angiotensin, activating factors VII and XI.
Genetic defects prolong whole-blood clotting time but do not cause clinical symptoms.
