Target Information
NISCH acts as a functional imidazoline-1 receptor (I1R) candidate or as a membrane-associated mediator of I1R signaling. It binds multiple imidazoline ligands that initiate cell-signaling cascades involved in cell survival, growth, and migration. Activation by rilmenidine increases phosphorylation of MAPK1 and MAPK3 in RVLM neurons, while blocking with efaroxan inhibits this effect.
NISCH modulates Rac-regulated signaling pathways, suppressing Rac1-stimulated cell migration via PAK1 inhibition and blocking Rac1-induced NF-κB and cyclin D1 activation. It also reduces LIMK1 Tyr-508 phosphorylation, inhibiting Rac-induced migration and invasion in epithelial cells. Overexpression inhibits lamellipodia formation and supports protection against apoptosis. Additionally, it associates with IRS4 to enhance insulin activation of MAPK1 and MAPK3, and induces redistribution of ITGA5 integrin to endosomal structures.
