Target Information
TRF1은 텔로미어 길이 유지의 음성 조절 인자로 알려져 있습니다. 최근 TRF1에 결합하는 Tankyrase와 Tin2 두 단백질이 확인되었습니다.
Tankyrase는 ankyrin 반복 서열 및 poly(ADP-ribose) polymerase (PARP) 촉매 도메인과 상동성을 가지며, TRF1을 통해 텔로미어에 위치합니다.
Tankyrase의 PARP 활성은 TRF1의 ADP-리보실화에 관여하며, 이는 TRF1의 텔로미어 DNA 결합 능력을 감소시킵니다.
TIN2는 TRF1과 상호작용하며 텔로미어 길이 조절에 필수적입니다. TRF1과 TIN2는 모두 광범위하게 발현되며, 텔로머라아제에 의한 텔로미어 연장을 억제하는 역할을 합니다.
다만, 내인성 TIN2는 발현량이 낮아 일부 세포 시스템에서 면역염색으로 관찰하기 어려울 수 있습니다.
