Product Specific Information
The monoclonal antibody mT2.4 inhibits murine TLR2-mediated cell activation. It masks the lipopeptide binding site in the recombinant extracellular domain of TLR2 (TLR2ECD) and does not hinder receptor complex formation. The antibody reacts with native TLR2 protein but not with denatured TLR2 protein. It stains both overexpressed and endogenous cell surface and intracellular mouse TLR2.
Target Information
TLR2 is a member of the Toll-like receptor (TLR) family, which plays a crucial role in pathogen recognition and activation of innate immunity. TLR proteins act through adaptor molecules such as MyD88 and TIRAP to activate kinases and transcription factors including PKC alpha/beta and NF-kappa-B.
TLR2 can form heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins. TLRs are highly conserved across species and exhibit distinct expression patterns. TLR2 is abundant in peripheral blood leukocytes and mediates host responses to Gram-positive bacteria and yeast via NF-kappaB activation.
Expression of TLR2 is modulated by immune response modifiers: LPS, GM-CSF, IL-1, and IL-10 upregulate TLR2, while IL-4, IFN-gamma, and TNF downregulate its expression in monocytes.
