Target Information
Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene. It is a key extracellular matrix component in cartilaginous tissue, providing resistance to compression.
The human aggrecan protein (~250 kDa, 2316 amino acids) can be expressed in multiple isoforms due to alternative splicing. It has a bottlebrush structure with chondroitin sulfate and keratan sulfate chains attached to a protein core.
Aggrecan synthesis and degradation are crucial in cartilage deterioration during joint injury, disease, and aging. It contains three interglobular domains (G1, G2, G3) involved in aggregation, hyaluronan binding, cell adhesion, and chondrocyte apoptosis.
Aggrecan fragments are released into synovial fluid during osteoarthritis progression due to cleavage by aggrecanase and MMP activity. Antibodies BC-3 and BC-4 detect neo-epitopes of Aggrecanase (ARGSVXXX) and MMP (XXXVDIPEN), respectively.
