Product Specific Information

The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L, also known as lymphocyte function-associated antigen-1 (LFA-1) alpha chain. LFA-1, formed by non-covalent association of CD11a with CD18 (integrin beta 2), functions as an adhesion molecule involved in lymphocyte and granulocyte activity. Ligands include CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3). CD11a is expressed on all leukocytes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Use in flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤ 0.4 µg/test (100 µL final volume). Cell number may vary from 10⁵ to 10⁸ cells/test. Titrate for optimal results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability testing.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails prior to combining with cells.
• For single-color controls, use 5 µL of buffer per 100 µL of cell sample (10³–10⁸ cells).

Note: Internal testing shows NovaFluor Yellow 610 may non-specifically stain B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 strains.

Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 forms the leukocyte function-associated antigen-1 (LFA-1) complex. CD11a mediates leukocyte adhesion via interactions with ICAMs 1–3 and plays key roles in lymphocyte activation and migration. It is expressed on leukocyte plasma membranes and transitions between low- and high-affinity conformations upon stimulation. CD11a dysfunction is linked to benign cephalic histiocytosis and leukocyte adhesion deficiency. Therapeutic targeting includes anti-CD11a antibodies (e.g., Efalizumab) and small molecules such as simvastatin that modulate immune activity.
