Product Specific Information

The M17/4 monoclonal antibody reacts with mouse CD11a, a 180 kDa integrin alpha L, also known as lymphocyte function-associated antigen-1 (LFA-1) alpha chain. LFA-1, formed by the non-covalent association of CD11a with CD18, plays a critical role in lymphocyte and granulocyte adhesion and function. CD11a is expressed by all leukocytes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤ 0.4 µg/test (100 µL final volume).
Cell count per test: 10⁵–10⁸ cells. Titrate antibody for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample or per 100 µL cell suspension.

Target Information

CD11a (LFA-1α, ITGAL) forms the αLβ2 integrin complex (LFA-1) with CD18. It mediates leukocyte adhesion via ICAMs 1–3 and participates in lymphocyte co-stimulatory signaling. CD11a transitions between low- and high-affinity conformations depending on cell activation, influencing immune cell adhesion, migration, and effector functions.

CD11a dysfunction is associated with:

• Benign cephalic histiocytosis
• Leukocyte adhesion deficiency

Therapeutic relevance includes:

• Anti-CD11a antibody Efalizumab (Raptiva) for inflammation control
• Simvastatin binding to CD11a allosteric site for immunomodulation
