Target Information
This single-chain zymogen (Factor XII) is converted to a two-chain serine protease consisting of a heavy chain (alpha-factor XIIa) and a light chain. The heavy chain includes fibronectin-type domains, EGF-like domains, a kringle domain, and a proline-rich domain, while the light chain contains a catalytic domain.
Upon activation, further cleavage of the heavy chain produces beta-factor XIIa. Prekallikrein is cleaved by factor XII to form kallikrein, which then processes factor XII to alpha- and beta-factor XIIa. Active factor XIIa participates in blood coagulation initiation, fibrinolysis, and bradykinin/angiotensin generation. It also activates coagulation factors VII and XI.
Defects in this gene do not cause clinical symptoms but prolong whole-blood clotting time.
