Product Specific Information

The APB5 monoclonal antibody reacts with the mouse CD140b molecule, the beta chain of the platelet-derived growth factor receptor (PDGFRB). PDGFRB is a receptor tyrosine kinase that forms dimers upon ligand binding and phosphorylates substrates. Dimers may be alpha/alpha, beta/beta, or alpha/beta, serving as a substrate for kinase activity. CD140b is expressed in embryonic tissues and mesenchymal-derived adult mouse cells, and plays a role in fibrous atherosclerotic lesion formation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on NIH/3T3 cells at ≤1.0 µg/test (100 µL sample volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Titrate for optimal performance.

Fluorochrome Information:
APC-eFluor 780 emits at 780 nm and is excited by a red laser (633–647 nm). Ensure instrument compatibility.

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be empirically determined.

Target Information

PDGFRB is a receptor tyrosine kinase implicated in atherogenesis and cell transformation by the SIS oncogene. It acts as a mitogen for mesenchymal-origin cells. Receptor composition (homodimer or heterodimer) is determined by the bound growth factor. The gene is located on chromosome 5 near GM-CSF and M-CSFR genes, all potentially involved in 5-q syndrome. A translocation between chromosomes 5 and 12 fusing PDGFRB with the ETV6 leukemia gene results in chronic myeloproliferative disorder with eosinophilia.
