Target Information
ATG3 (APG3) is a conjugating enzyme required for APG8 lipidation, a key step in autophagy initiation. Autophagy is a process of bulk degradation of cellular proteins via the autophagosomal-lysosomal pathway, essential for normal growth control and often defective in tumor cells. It helps preserve cellular nutrients under starvation and maintains turnover of cytosolic components. This process is negatively regulated by TOR through phosphorylation of APG1. ATG3 acts as an E2-like enzyme during autophagosome formation by catalyzing the Atg8-phosphatidylethanolamine (Atg8-PE) conjugate formation, critical for autophagy.
