Target Information
Tryptase gamma, also known as serine protease 31 or transmembrane tryptase, is an enzyme encoded by the TPSG1 gene in humans. Tryptases belong to the trypsin-like serine protease family (peptidase family S1). They are enzymatically active only as heparin-stabilized tetramers and resistant to endogenous proteinase inhibitors.
Several tryptase genes are clustered on chromosome 16p13.3, with four functional genes identified: alpha I, beta I, beta II, and gamma I. Each has allelic variants (alpha II, beta III, gamma II). Beta tryptases are the main isoenzymes in mast cells, while alpha-tryptases predominate in basophils.
TPSG1 differs from other tryptase genes by having a C-terminal hydrophobic domain that may act as a membrane anchor. Tryptases are implicated in asthma and other allergic or inflammatory disorders.
