iNOS (Inducible Nitric Oxide Synthase, NOS) is an enzyme producing nitric oxide (NO), a gaseous free radical acting as a signaling molecule in vasorelaxation, neurotransmission, and cytotoxicity. iNOS is expressed in the liver and inducible by lipopolysaccharide and cytokines.
NOS catalyzes the oxidation of L-arginine to produce L-citrulline and NO. There are three isoforms: neuronal NOS (nNOS, type I), endothelial NOS (eNOS, type III), and inducible NOS (iNOS, type II).
All NOS isoforms contain calmodulin, NADPH, FAD, and FMN binding domains. iNOS is found in macrophages, hepatocytes, synoviocytes, and smooth muscle cells. Cytokines such as IFN-γ, TNF, IL-1, IL-2, and LPS increase iNOS expression and activity.
After induction, iNOS shows delayed activation followed by sustained NO production. iNOS pseudogenes are located within the Smith-Magenis syndrome region on chromosome 17. Dysfunction of iNOS is associated with diseases such as achalasia and impotence.
