Product Specific Information

The 4B12 monoclonal antibody reacts with mouse CCR7 (CD197), a chemokine receptor for CCL19 and CCL21. CCR7 mediates lymphocyte migration to secondary lymphoid organs and is involved in T cell, B cell, and dendritic cell localization during immune responses.

CCR7 also serves as a marker for central memory T cells (TCM) and plays roles in thymocyte development. Deficiency in CCR7 leads to abnormal thymic architecture and impaired T cell development.

Applications

  • Reported for flow cytometric analysis.
  • Tested using mouse splenocytes and thymocytes.
  • Recommended ≤1 µg/test (100 µL volume, 10⁵–10⁸ cells/test).

  • Alexa Fluor® 700: Emission 723 nm, Excitation 633–647 nm
  • Compatible with Red Laser (He-Ne 633 nm)
  • Recommended filter: 685 LP mirror and 710/20 filter

Staining Protocol (4B12)

  1. Prepare cell suspension and block FcγIII/II receptors with 5 µg/million cells of anti-mouse CD16/32 (Product #14-0161) for 15 minutes on ice.
  2. Without washing, add 1 µg/million cells of 4B12 antibody and incubate at 37°C (recommended) or 2–8°C for 30 minutes.
  3. Wash once with 3 mL of Flow Cytometry Staining Buffer (Product #00-4222) and decant.
  4. Analyze cells on flow cytometer or proceed with secondary staining on ice.

Note:

  • Staining at 37°C enhances brightness and population resolution.
  • Co-staining with CCL19-Fc (Product #14-1972) may be difficult due to binding condition differences and steric hindrance.
  • 4B12 binding can prevent CCL19-Fc binding, but not vice versa.

Filtration: 0.2 µm post-manufacturing filtered.


Target Information

CCR7 is a G protein-coupled chemokine receptor induced by Epstein-Barr virus (EBV). It is expressed in blood, bone marrow, lymph nodes, and intestines, particularly in lymphoid tissues and activated B/T lymphocytes. CCR7 regulates memory T cell migration and dendritic cell maturation. Its specific ligand is CCL19 (ECL).

Receptors in the C-C chemokine family include CCR1–CCR5 and the Duffy antigen, which are critical for T cell chemotaxis and phagocyte migration to inflammation sites.
