This antibody reacts with an extracellular epitope of CD11a (alpha subunit of human LFA-1), a 170–180 kDa type I transmembrane glycoprotein expressed on B and T lymphocytes, monocytes, macrophages, neutrophils, basophils, and eosinophils.

Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 constitute leukocyte function-associated antigen 1 (LFA-1), the αLβ2 integrin. CD11a plays a central role in leukocyte intercellular adhesion through interactions with ICAMs 1–3 and functions in lymphocyte costimulatory signaling.

It is expressed on leukocyte plasma membranes in a low-affinity conformation. Upon stimulation by chemokines or other signals, CD11a shifts to a high-affinity form that supports tight binding to ICAM ligands. CD11a is involved in immune cell interaction, tissue-specific migration, differentiation, proliferation, and T-cell effector functions.

Blocking CD11a function using specific antibodies or small molecules is a therapeutic approach for inflammatory diseases. For example:

• Efalizumab (Raptiva): a humanized anti-CD11a antibody used to inhibit T-cell migration.
• Simvastatin: binds to CD11a allosteric site, leading to immunomodulation and increased lymphocytic cholinergic activity.

Two transcript variants encoding different isoforms of CD11a have been identified. Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
