Product Specific Information

Description:
The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and virtually all mature T cells, and is upregulated upon T-cell stimulation. CD27 binds to CD70, playing an important role in T cell–B cell interaction.

Applications Reported:
This O323 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.25 µg) per test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm and can be excited with the red laser (633 nm).
Recommended filter set: 685 LP mirror and 710/20 filter.
Excitation: 633–647 nm; Emission: 723 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD27 is a 50 kDa member of the tumor necrosis factor (TNF) receptor superfamily that includes CD40 and CD30. This family regulates cell proliferation and death. CD27 and its ligand CD70 are mainly expressed in lymphocytes. High CD27 expression depends on proper antigen receptor ligation, while CD70 requires co-stimulatory or pro-inflammatory signals.

CD27 is a disulfide-linked homodimer expressed on mature thymocytes, peripheral blood T cells, and a subset of B cells. T-cell activation via the TCR-CD3 complex increases CD27 expression and releases soluble CD27 (sCD27, 28–32 kDa), detectable in plasma, urine, or spinal fluid. Soluble CD27 serves as a prognostic marker for acute and chronic B cell malignancies.

RgpA, a cysteine proteinase, can degrade CD27 and counteract T cell activation mediated by CD27–CD70 interaction. CD27-binding protein (SIVA) is a proapoptotic protein associated with CD27-mediated apoptosis.
Diseases linked to CD27 dysfunction include Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
