Product Specific Information

Description:
The WM-53 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein of the Siglec family. CD33 is expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of lysed whole blood.
Recommended use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm and can be excited with the red laser (633 nm).
Typical setup: 685 LP mirror and 710/20 filter.
Excitation: 633–647 nm; Emission: 723 nm; Laser: Red.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD33 is a sialic acid-binding immunoglobulin-like lectin (Siglec) family transmembrane protein containing ITIM motifs. It recruits SHP-1 and SHP-2 phosphatases and is involved in receptor regulation via ubiquitin-mediated removal.
Expressed on myelomonocytic lineage cells, CD33 binds sialic acid residues on cell surfaces and serves as a therapeutic target for acute myeloid leukemia.
Found on granulocyte and macrophage precursors but not pluripotent stem cells.
Useful marker for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Associated diseases include gallbladder lymphoma and extracutaneous mastocytoma.
