Target Information
Tumor necrosis factor (TNF)-induced signaling occurs via association of TNF receptor (TNFR) with adaptor proteins such as TRAFs. TRAFs are cytoplasmic adapter proteins mediating signal transduction from TNF-receptor superfamily members (e.g., RANK, CD30, CD40) and the interleukin-1 receptor. The carboxy-terminal region of TRAFs is required for self-association and receptor interaction following ligand-induced oligomerization. Six TRAF members (TRAF1–TRAF6) have been identified. TRAF5 is a 558-amino-acid protein involved in NF-kB and c-Jun N-terminal kinase/stress-activated protein kinase activation by TNF receptor superfamily members, including CD27, CD30, CD40, and lymphotoxin-beta receptor. TRAF5 gene disruption causes defects in CD40-CD27 mediated lymphocyte activation.
