This product is diluted and provided in a ready-to-use formulation.
A recommended positive control tissue is Hodgkin lymphoma, though other positive controls may be used.
The antibody is intended for professional laboratory use in detecting Fascin protein in formalin-fixed, paraffin-embedded tissues using manual qualitative immunohistochemistry (IHC). It should be applied after tumor diagnosis by conventional histopathology.
Fascin Function:
Human Fascin is an actin-bundling protein localized to microspikes and stress fibers, involved in microfilament bundle formation. It is primarily expressed in dendritic cells, while lymphoid, myeloid, and plasma cells are negative. Reed-Sternberg cells in Hodgkin’s lymphoma are positive for Fascin staining. Epstein-Barr virus may induce Fascin expression in B cells. Clone 55K-2 serves as an effective marker for Reed-Sternberg cells.
Usage Recommendations:
- Use with formalin-fixed, paraffin-embedded sections.
- Pretreat deparaffinized tissue with heat-induced or enzymatic epitope retrieval.
- Follow standard IHC staining procedures: primary antibody → link antibody → enzyme complex → chromogenic substrate.
- Interpret results using a light microscope.
Control Tissues:
- Run a positive tissue control with every staining procedure.
- Positive control tissue should include both positive and negative staining cells.
- Use fresh, well-characterized specimens for external controls.
- Weak positive tissues are recommended to detect subtle changes in antibody sensitivity.
- Negative controls (internal or external) should confirm absence of nonspecific staining.
Target Information
Fascin organizes filamentous actin into bundles with a 1:1 actin/fascin ratio and is involved in the assembly of actin filament bundles in microspikes, membrane ruffles, and stress fibers.
