Product Specific Information
- Recommended positive control tissue: Bone marrow (other tissues also applicable).
- Intended for detection of Erythropoietin (EPO) in formalin-fixed, paraffin-embedded tissue using manual qualitative IHC.
- Recognizes a ~37 kDa protein identified as EPO, a secreted glycosylated cytokine regulating erythropoiesis.
- EPO is synthesized in the kidney and stimulates proliferation and maturation of bone marrow erythroid precursor cells.
- Found in plasma, promoting erythroid differentiation and hemoglobin synthesis.
- Pretreatment with heat-induced or enzymatic epitope retrieval is recommended.
- IHC staining involves sequential application of primary antibody, secondary antibody, enzyme complex, and chromogen substrate, with washing steps.
- Positive and negative control tissues must be included in each staining procedure.
- Positive control tissues should exhibit weak positive staining for optimal quality control.
- Negative controls should confirm absence of specific staining and minimal background.
Target Information
Erythropoietin (EPO) is a growth factor essential for survival, proliferation, and differentiation of erythroid precursors. It promotes final maturation and hemoglobin synthesis. EPOR expression is high in precursor cells and decreases with maturation.
In fetuses, hepatocytes are the main source of EPO; in adults, renal interstitial cells are predominant. EPO expression increases under hypoxia and has been detected at low levels in lung and brain tissue.
Recent studies suggest EPO may play a role in neuronal protection and repair.
