Target Information
The G-protein-coupled receptor (GPCR) superfamily is comprised of an estimated 600–1,000 members and represents the largest known class of molecular targets with therapeutic relevance.
The nociceptin receptor is a G-protein coupled opioid receptor that binds the endogenous neuropeptide nociceptin. Ligand binding induces conformational changes that trigger G protein-mediated signaling, inhibiting adenylate cyclase and calcium channel activity. Arrestins modulate G protein signaling and activate alternative pathways, including MAP kinase activation.
This receptor plays a key role in modulating nociception, pain perception, and locomotor activity regulation by nociceptin.
