Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either N- or C-terminal regions of recombinant proteins to enable isolation or purification by immobilized metal affinity chromatography (IMAC).
Epitope tagging allows detection of recombinant proteins for which no specific antibody is available. His-tagged proteins can be purified using Ni²⁺-NTA adsorbent due to strong and selective chelation of histidine clusters to nickel ions.
His tag-specific antibodies, such as clone 2D5, are used to facilitate detection or co-immunoprecipitation of tagged proteins.
