Target Information
Furin is a membrane-associated, calcium-dependent serine protease belonging to the subtilisin-like prohormone convertase (PC) family. These enzymes cleave prohormones and neuropeptide precursors, as well as various cellular, viral, and bacterial proteins transported via the constitutive secretory pathway.
Furin and related PCs share structural features including a ~10 kDa N-terminal proregion, a ~55 kDa catalytic domain, and a variable C-terminal domain. Activation occurs after proregion cleavage within specific cellular compartments. Furin uniquely possesses a transmembrane domain and cleaves at the carboxyl terminus of the consensus sequence RX(K/R)R.
The acidic sequence C771PSDSEEDEG780 localizes Furin to the trans-Golgi network, and phosphorylation of serine residues in this region regulates intracellular routing. The Y759KGL762 motif mediates internalization from the cell surface.
