The NCF4 gene encodes a cytosolic regulatory component of the superoxide-producing phagocyte NADPH-oxidase, a multicomponent enzyme system crucial for host defense.
This protein is preferentially expressed in myeloid lineage cells and interacts with NCF2 (p67-phox) and NCF1 (p47-phox), forming a complex that associates with RAC1 and translocates to the membrane upon cell stimulation.
The PX domain binds phospholipid products of PI(3) kinase, indicating involvement in PI(3) kinase-mediated signaling.
Phosphorylation negatively regulates enzyme activity.
Alternatively spliced transcript variants encoding distinct isoforms have been identified.
