Product Specific Information

The HIB19 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed on B cells throughout development, excluding terminally differentiated plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, Leu13, and MHC class II, associating with the BCR to mediate signaling that induces tyrosine phosphorylation, calcium flux, and B cell proliferation. The SJ25C1 and HIB19 antibodies recognize overlapping epitopes.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (1.0 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• The conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance.
• Add 5 µL of CellBlox Plus per sample or per 100 µL of cell suspension for optimal results.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on all peripheral B cells and certain myeloid leukemia cells. CD19 acts as a co-receptor with BCR, lowering the activation threshold for antigen-dependent stimulation and serving as a central regulator of B cell signaling. Mutations in CD19 can lead to hypogammaglobulinemia, while overexpression is associated with B cell hyperactivity.
