Product Specific Information

The 2C9.G3 (HMb3-1) monoclonal antibody reacts with mouse and rat CD61 (integrin beta3), expressed on activated T cells, granulocytes, and platelets. CD61 forms heterodimers with integrin alpha subunits:

• alphaV (CD51) → Vitronectin Receptor
• alphaIIb (CD41) → gpIIb/IIIa complex

These complexes mediate adhesion to extracellular matrix components such as fibrinogen, fibronectin, vitronectin, thrombospondin, and von Willebrand factor.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes and bone marrow cells (≤ 0.1 µg/test)

A test is defined as the amount of antibody staining a cell sample in a final volume of 100 µL. Recommended careful titration for optimal performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.

Based on Phiton™ nucleic acid dye structures engineered for reduced spillover and spread impacts.

Optical Properties:

• Laser: 561 nm (Yellow)

Note: NovaFluor Yellow 730 may non-specifically stain B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 mice.

Target Information

CD61 (GPIIIa, ITGB3) is a 105 kDa glycoprotein found on megakaryocytes, platelets, and their precursors. It functions as a receptor for fibrinogen, fibronectin, von Willebrand factor, vitronectin, and thrombospondin.

• Participates in cell adhesion and signal transduction
• Expressed on platelets, endothelial cells, monocytes, and osteoclasts
