Target Information
PSD-93 (also known as chapsyn-110) is a plasma membrane-associated protein found in synaptic junctions. It is uniquely expressed in Purkinje neuron cell bodies and dendrites. PSD-93 contains three PDZ domains (~90 amino acids each), one SH3 domain, and a carboxyl-terminal guanylate kinase homology (GuK) domain that is enzymatically inactive.
PDZ-domain interactions are thought to play a role in receptor and channel clustering, contributing to neuronal plasticity. PSD-93 is believed to cluster NMDA receptors and shaker-type potassium channels at the synaptic membrane.
NMDA receptors and shaker-type potassium channels share a C-terminal T/SXV motif that interacts with PSD-93 PDZ domains. Additionally, neuronal nitric oxide synthase (nNOS), which lacks this motif but has its own PDZ domain, can associate with PSD-93 through PDZ–PDZ interactions.
