Product Specific Information

The RM2-5 monoclonal antibody reacts with the mouse CD2 molecule, an approximately 50–55 kDa cell surface receptor expressed by all mouse lymphocytes. In mice, CD2 is expressed on B lineage cells from the pre-B cell stage to mature B cells and acts as a ligand for CD48, playing roles in adhesion and T cell activation.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Tested on mouse splenocytes and bone marrow cells. Recommended ≤0.2 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.
• Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample or per 100 µL of cell suspension (10³–10⁸ cells).
  • Add buffer to antibody cocktails before mixing with cells.

Note: Internal testing shows non-specific staining of B cells in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 strains. Other strains not tested.

Target Information

CD2 (LFA-2) is a monomeric surface antigen (45–58 kDa) expressed on human T lymphocytes and some NK cells, but not on B cells. It mediates adhesion via interaction with CD58 and is involved in T-cell activation. CD2 deficiency can reduce intestinal inflammation and affect immune responses. Dysregulation is associated with certain carcinomas and immune deficiencies.
