Target Information
Epithelial sodium channels (ENaC) are amiloride-sensitive members of the degenerin/epithelial sodium channel (Deg/ENaC) superfamily of ion channels. These channels share structural similarities, including two intracellular termini, two membrane-spanning segments, and a large extracellular loop with a conserved cysteine-rich region.
ENaC consists of three homologous isoforms: alpha, beta, and gamma. It plays a key role in sodium and fluid balance in the kidney, lung, and colon, and mediates aldosterone-dependent sodium reabsorption in the distal nephron, thus regulating blood pressure.
Mutations in ENaC subunits are linked to disorders such as Liddle’s syndrome (gain-of-function) and pseudohypoaldosteronism type I (loss-of-function).
