Anti-sperm antibodies have been correlated with cases of unexplained infertility, suggesting a role in blocking fertilization. The identification and characterization of sperm antigens are important for understanding immunologic infertility and developing targeted contraception.

The protein encoded by the SPAG1 gene is recognized by anti-sperm agglutinating antibodies from an infertile woman. Immunization of female rats with recombinant human SPAG1 protein reduced fertility. This protein localizes to the plasma membrane of germ cells in the testis and to the post-acrosomal plasma membrane of mature spermatozoa.

Recombinant SPAG1 binds GTP and exhibits GTPase activity, indicating a role in GTP signal transduction pathways involved in spermatogenesis and fertilization. Two transcript variants encode the same protein.
