Target Information
Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. It acts as a ligand for PD-1 along with PD-L1 and plays an important role in immune checkpoint regulation. PD-L2 is primarily expressed in dendritic cells but can be induced in various cell types depending on the microenvironment.
PD-L2 expression is upregulated by cytokines such as IL-4, TNF-alpha, and IFN-gamma. Compared to PD-L1, PD-L2 shows higher affinity to PD-1 (2–6×). It contributes to immune homeostasis by inhibiting T cell activation and cytokine secretion.
Recent studies revealed PD-L2 also binds to RGMb, regulating respiratory tolerance through BMP/neogenin signaling pathways. The human PD-L2 monomer consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
