Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, regulating G1 or G2/M arrest. Structurally, p53 consists of an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is crucial for p53 activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 also plays roles in aging, oxidative stress, and learning-related processes. Normally expressed at low levels, p53 is highly expressed in transformed cell lines, contributing to malignancy. Mutations in TP53 disrupt DNA binding and tumor suppressor function, occurring both somatically and germline (e.g., Li-Fraumeni syndrome).
