Target Information
The second messengers cAMP and cGMP are key regulatory molecules involved in a wide variety of signal transduction pathways. Their levels are controlled by synthesis via nucleotide cyclases and degradation via cyclic nucleotide phosphodiesterases (PDEs).
PDE6 acts as the effector enzyme in the G protein-mediated signal transduction cascade of the visual system. It consists of rod and cone-specific catalytic subunits (α′, α, and β), an inhibitory subunit (γ), and a δ subunit of unknown function.
The catalytic core forms α′/α′ homodimers in cones and α/β heterodimers in rods. The C-terminus of both catalytic and inhibitory subunits is modified by methylation, myristoylation, and prenylation — modifications essential for complex assembly and membrane association.
