Target Information

Receptor-tyrosine-kinase-like orphan receptor 1 (ROR1) is a tumor-associated surface protein predominantly expressed during embryogenesis, where it plays roles in organ morphogenesis, nervous system development, and neural progenitor cell maintenance. It is virtually absent from normal adult tissues except in immature B cell precursors (hematogones) and adipocytes. ROR1 is notably overexpressed in several malignancies, including chronic lymphocytic leukemia (CLL), mantle cell lymphoma (MCL), acute lymphoblastic leukemia (ALL), marginal zone lymphoma, and lung adenocarcinoma.

Initially identified in neuroblastoma cell lines as an orphan receptor tyrosine kinase, ROR1 and its homolog ROR2 were named for their unknown ligands. Wnt-5a has since been proposed as a ligand, and ROR1 may act as a pseudokinase, promoting proliferation and resistance to apoptosis in cancer cells through Wnt-5a interaction and TCL1-mediated AKT activation. ROR1 is expressed as a glycoprotein with extracellular Ig-like, Frizzled, and Kringle domains, and an intracellular tyrosine kinase domain.
