Product Specific Information
The monoclonal antibody A1exF5 recognizes both human and mouse Arginase 1, a cytosolic enzyme (Arg1).
This clone is compatible with standard intracellular staining protocols and the Foxp3/Transcription Factor Staining Buffer Set.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometry using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
Refer to BestProtocols®: Protocol A for cytoplasmic intracellular proteins.
Use ≤0.5 µg per test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Antibody titration recommended for optimal performance.
Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer ([Product # 00-8222]) or 1-step Fix/Lyse Solution ([Product # 00-5333-54]) for up to 3 days at 4°C in the dark with minimal effect on brightness and FRET efficiency.
Excitation: 488–561 nm
Emission: 775 nm

Target Information
Arginase-1 (Arg1) is a 35 kDa enzyme converting L-arginine to urea and L-ornithine, the final step in the urea cycle.
It regulates nitric oxide production by depriving NO synthase of its substrate.
Arginase 1 is expressed in liver, neutrophils, myeloid-derived suppressor cells (MDSC), and neural stem cells.
In mice, it is a hallmark of alternatively activated macrophages (M2a).
Defects in Arginase 1 cause argininemia, an autosomal recessive disorder characterized by hyperammonemia.
