Product Specific Information

  • For Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly during late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that mediates association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.

  • Ki-67 shows cell cycle-dependent localization:
    • Perinucleolar expression at G1
    • Nuclear matrix expression at G2
    • Chromosomal expression during M phase
  • Commonly used as a proliferation marker since it is absent in G0 cells and increases from G1 through mitosis.
  • Useful for determining the cell growth fraction in neoplasms.
  • Correlates with tumor grade; low Ki-67 index indicates low-grade tumors.
  • Routinely used as a neuronal marker of cell cycling and proliferation.
