Target Information
RNA polymerase III synthesizes tRNA, 5S rRNA, 7SL RNA, and U6 snRNA, and is overexpressed in many transformed cell lines and tumors. This high expression supports rapid cell growth and division. The TFIIIC2 complex, composed of TFIIIC220, TFIIIC110, TFIIIC102, TFIIIC90, and TFIIIC63, recruits RNA polymerase III and TFIIIB to promoter elements and plays a key role in malignant cell deregulation. TFIIIB includes TBP, BRF1, and TFIIIB itself, and its expression is upregulated in transformed cells. Tumor suppressors such as RB and p53 are often inactivated in carcinomas, reducing their ability to bind and inhibit TFIIIB function.
