Product Specific Information

This KIKSI0803 monoclonal antibody recognizes STAT1 (Signal Transducer and Activator of Transcription 1) when phosphorylated on tyrosine 701. STAT proteins are activated by receptor-associated Janus kinases (JAKs) upon ligand binding. Once phosphorylated, STAT1 translocates to the nucleus to regulate gene transcription in a cytokine- and cell-type–specific manner.

Phosphorylation of STAT1 on Tyr701 by JAK1 and JAK2 is essential for dimerization, nuclear translocation, and DNA binding. Upon IFN-γ stimulation, STAT1 forms homodimers or heterodimers with STAT3, binding to GAS promoter elements. Under IFN-α or IFN-β stimulation, STAT1 forms heterodimers with STAT2, binding to ISRE promoter elements.

Specificity of clone KIKSI0803 was verified by ELISA, flow cytometry, and western blotting.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining of human peripheral blood cells. Use at 5 µL (0.06 µg) per 100 µL test volume. Recommended cell range: 10⁵–10⁸ cells/test.

Staining Protocol:
Use Protocol C (Two-step: Fixation/Methanol).
Protocol A (cytoplasmic) and Protocol B (nuclear) are not suitable.

Target Information

STAT1 is a transcription factor activated by interferons (α, β, γ), EGF, PDGF, and IL-6. It regulates genes involved in cell growth, apoptosis, immune response, and lipid metabolism. STAT1 activation by JAK kinases enables nuclear translocation and transcriptional regulation of multiple genes. The STAT1 gene is located on chromosome 2 and exists in two alternatively spliced isoforms. STAT1, STAT3, STAT5a, and STAT5b have broad activation profiles in response to cytokines and growth factors.
