Target Information

Histone H2A.X (H2AX) is a member of the histone H2A family, one of the four core histones forming the nucleosome core particle. In eukaryotes, DNA double-strand breaks (DSBs) trigger phosphorylation of serine 139 at the carboxy terminus of histone H2AX, resulting in gamma-H2AX formation. This phosphorylation can be detected by Western blotting or immunofluorescence, reflecting DSB frequency. Phosphatidylinositol 3-kinases are implicated in H2AX phosphorylation, though it remains unclear whether ATM is the primary kinase or if DNA-PK and ATR also contribute.

Structurally, H2A.X contains 143 amino acids and is synthesized during both G1 and S phases. Its mRNA includes polyA addition motifs and a polyA tail, along with conserved stem-loop and U7 binding sequences that regulate replication-type histone mRNA stability. Two forms of H2A.X mRNA exist: a 1600-base polyA-containing form and a 575-base non-polyA form. The short form behaves as replication-type, while the longer behaves as basal-type histone mRNA. Histone H2A.X maps to the 11q23.2–q23.3 region of the human chromosome and contributes to chromatin structure and DNA organization. The H2A.X variant specifically regulates MDC1 interaction at DNA damage sites.
