Target Information
In mammals, several isoforms of p21-Activated Protein Kinases (PAKs) are identified:
- α-PAK (PAK1) and β-PAK (PAK3) are mostly brain-specific.
- γ-PAK (PAK2) is expressed ubiquitously (Jakobi et al., 2003).
Mutations in the gene encoding PAK3 are linked to X-linked mental retardation, and studies suggest PAK3 is a key regulator of synapse formation and plasticity in the hippocampus (Boda et al., 2004).
PAK3 also plays an important role in regulating cell shape, motility, and apoptosis (Jakobi et al., 2003; Walter et al., 1998).
Autophosphorylation at Thr402 is essential for PAK activation (Jakobi et al., 2000).
