Target Information
The antimicrobial protein BPI is a 55 kDa protein found in the primary (azurophilic) granules of human neutrophils and also detected on the surface of neutrophils, small intestinal, and oral epithelial cells.
BPI is a bactericidal compound present in polymorphonuclear cells (PMN) and at lower levels in eosinophils. It exhibits high affinity for the lipid A region of lipopolysaccharides (LPS) in gram-negative bacteria, leading to multiple anti-infective activities including:
- Cytotoxicity via sequential damage to bacterial membranes
- Neutralization of gram-negative bacterial LPS
- Opsonization to enhance phagocytosis by neutrophils
BPI may play a critical role in LPS-triggered airway disease, as airway epithelial cells express the BPI gene and produce the protein. Inflammation induced by LPS may contribute to rapid airflow decline, a severe complication of hematopoietic cell transplantation.
Additionally, a 21 kDa recombinant fragment of BPI (rBPI21) confers survival advantage against invasive pneumococcal disease by binding to pneumolysin from gram-positive bacteria.
