Product Specific Information

Description:
The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding mature erythrocytes and platelets. It plays a crucial role in lymphocyte activation through its tyrosine phosphatase enzymatic activity.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Validated for flow cytometric analysis of mouse bone marrow cells at ≤0.4 µg/test.
A test is defined as staining a cell sample in 100 µL final volume (10⁵–10⁸ cells/test).
Antibody titration is recommended for optimal performance.

NovaFluor Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer:
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample before combining with cells.
For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins).
It regulates Src-family kinases, T/B-cell antigen receptor signaling, and cytokine receptor signaling.
CD45 isoforms arise via alternative splicing and differ in extracellular domains.
It also modulates integrin-mediated signaling, apoptosis, and immune functions.
