Product Specific Information
The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed on all hematopoietic cells except mature erythrocytes and platelets. The cytoplasmic domain has tyrosine phosphatase activity, essential for lymphocyte activation.
- Applications Reported: Flow cytometric analysis.
- Applications Tested: Tested on mouse bone marrow cells; use ≤ 0.2 µg per test (100 µL final volume).
Optimal antibody concentration should be titrated per assay.
Note: NovaFluor Yellow 590 shows strain-dependent non-specific staining in B cells of SJL mice, but not in BALB/c or C57BL/6 strains.
Compatibility and Usage Notes
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- Use CellBlox Blocking Buffer (Cat. B001T06F01) with all NovaFluor conjugates:
- Add 5 µL per sample (10³–10⁸ cells) before combining with cells.
- For antibody cocktails, mix buffer before adding to cells.

Target Information
CD45 (LCA, Leukocyte Common Antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It regulates Src-family kinases, cytokine receptor signaling, and lymphocyte activation.
CD45 exists in multiple isoforms (e.g., CD45RA) via alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
It plays key roles in T/B cell receptor signaling, apoptosis regulation, and integrin-mediated survival pathways.
