Target Information
Tumor necrosis factor (TNF) receptor-associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to various signaling pathways, playing key roles in regulating cell death and stress responses.
TRAF proteins possess a conserved domain that mediates interactions with cell receptors and other TRAFs, triggering distinct signaling cascades. TRAF2 and TRAF3 bind directly to CD40, a receptor crucial for NF-κB activation in B lymphocytes. TRAF2 also participates in CD40 signaling in nonhematopoietic cells and interacts with lymphotoxin-beta receptor (LTβR) along with TRAF3 and apoptosis regulators such as cIAP1 and Smac.
