Target Information
AHR (Ah Receptor)는 ARNT(HIF-1 Beta) 및 Drosophila 단백질 PER, SIM과 같은 단백질 패밀리에 속합니다. AHR은 약 200개의 아미노산으로 구성된 N-말단 PAS 도메인을 포함하며, 다양한 조직에서 발견됩니다.
AHR은 특정 DNA enhancer 서열에 결합하여 cytochrome P-450 (CYPIA1) 유전자의 mRNA 전사를 유도합니다. 이 유전자는 평면 방향족 탄화수소에 대한 생물학적 반응을 조절하는 리간드 활성화 전사 인자를 암호화합니다.
AHR은 xenobiotic 대사 효소인 cytochrome P450을 조절하며, 그 리간드는 방향족 탄화수소류입니다. AHR은 다양한 척추동물에서 발견되는 helix/loop/helix 형태의 리간드 활성화 전사 인자입니다. 알려진 리간드는 polycyclic 및 halogenated aromatic compounds(예: TCDD)입니다.
비활성 상태에서 AHR은 HSP90과 복합체를 이루어 세포질에 존재하며, 리간드 결합 후 핵 내 DNA 결합 형태로 변환되어 Arnt와 이합체를 형성합니다.
AHR 관련 질환으로는 eosinophilic fasciitis 및 seborrheic dermatitis가 보고되어 있습니다.
