Description:
The CBRM1/5 monoclonal antibody reacts with an activation-specific epitope of human Mac-1. It binds a subset of Mac-1 molecules on neutrophils and monocytes after stimulation but not on resting myeloid cells. Mac-1 participates in adhesive cell interactions, and the epitope recognized by CBRM1/5 localizes near the ligand-binding site on the alpha chain of Mac-1. This antibody blocks Mac-1 dependent adhesion to fibrinogen and ICAM-1 and inhibits chemoattractant-stimulated adhesion of eosinophils to ICAM-1.

Low-level activation may occur during blood processing; thus, some binding may appear in unstimulated samples, though higher expression is observed in activated cells.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometry on stimulated human peripheral blood cells. Use 5 µL (0.06 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal loss of signal. Clone-specific performance should be verified empirically.

Target Information

CD11b (Integrin alpha-M, ITGAM):
A 165 kDa adhesion molecule forming a heterodimer with integrin beta-2 (CD18), known as Mac-1 or CR3. Expressed on monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK and dendritic cells, and microglia. CD11b/CD18 mediates binding to ICAM-1, ICAM-2, ICAM-4, CD14, fibrinogen, and other ligands, playing key roles in cell adhesion and immune responses.

Expression varies by cell type and activation state:

• 8% of spleen cells
• 44% of bone marrow cells
• <1% of thymocytes

Commonly used as a microglial marker. Altered CD11b function is associated with systemic lupus erythematosus and ITGAM-related immune disorders.
