Target Information
Phosphoinositide-specific phospholipase C (PLC) plays a crucial role in receptor-mediated signal transduction by generating two second messengers, inositol 1,4,5-triphosphate and diacylglycerol, from phosphatidylinositol 4,5-bisphosphate. Mammalian PLC isozymes include PLCβ1–4, PLCγ1–2, PLCδ1–2, and PLCε.
PLCδ1 acts as a calcium signal amplifier and is activated by atypical GTP-binding proteins. It functions as an effector for transglutaminase II-mediated oxytocin receptor and α1B-adrenoreceptor signaling. Mouse PLCδ1 is highly expressed in brain, heart, lung, and testis.
Abnormal accumulation of PLCδ is observed in Alzheimer’s disease brains, suggesting a potential role in AD pathology. PLCδ2 is markedly expressed in type II intestinal metaplasia and adenocarcinoma, and its expression may serve as a marker for gastric malignant transformation.
