Target Information
Mammalian mitochondrial ribosomal proteins are encoded by nuclear genes and assist in protein synthesis within mitochondria. Mitochondrial ribosomes (mitoribosomes) consist of a small 28S subunit and a large 39S subunit, with an approximate 75% protein to rRNA ratio—opposite to prokaryotic ribosomes. Unlike prokaryotic ribosomes, mammalian mitoribosomes lack a 5S rRNA. Protein composition varies among species, making sequence homology recognition difficult.
The DAP3 gene encodes a 28S subunit protein involved in apoptotic pathways initiated by TNF-α, Fas ligand, and interferon-γ. It may bind ATP/GTP and function with mitoribosomal protein S27. Splice variants differ in the 5′ UTR but encode the same protein. Pseudogenes exist on chromosomes 1q and 2q.
