Target Information
CHMP4A는 endosomal sorting required for transport complex III (ESCRT-III)의 핵심 구성 요소로, multivesicular bodies(MVBs) 형성과 내재 소포(intraluminal vesicles, ILVs)의 생성에 관여합니다.
MVB는 내재 소포를 포함하며, 이들은 내막의 함입 및 절단을 통해 형성되어 리소좀으로 전달되어 막단백질, 성장인자 수용체, 리소좀 효소 및 지질의 분해를 가능하게 합니다.
ESCRT 기계는 ESCRT-0, -I, -II, -III 복합체의 순차적 작용을 필요로 하며, ESCRT-III 단백질은 ILV가 방출되기 전에 막으로부터 해리됩니다.
또한 ESCRT 복합체는 세포질분열의 최종 단계나 HIV-1과 같은 피막형 바이러스의 출아(budding) 과정에서도 막 절단에 관여합니다.
CHMP4A는 PDCD6IP와의 상호작용을 통해 HIV-1 p6 및 p9 의존적 바이러스 방출에 관여하며, 과발현 시 막 결합형 원형 배열을 형성하여 음의 곡률 및 외향성 출아를 촉진할 수 있습니다.
