Specificity Comments:
FAP (fibroblast activation protein) is a cell surface glycoprotein and serine protease expressed primarily in fetal mesenchymal tissues and epithelial cancer fibroblasts. In cancer, FAP promotes cellular proliferation, while in embryonic development it contributes to tissue remodeling. FAP acts as an integral membrane gelatinase composed of N-glycosylated, proteolytically inactive subunits.
FAP expression on chondrocyte membranes is upregulated by cytokines IL-1 and OSM, increasing in osteoarthritic patients. This expression is colocalized with MMP-1, MMP-13, and CD44 (variants v3 and v7/8). Mice lacking all copies of the FAP gene are fertile and show no developmental defects or altered cancer susceptibility.
Target Information
The protein encoded by this gene is a homodimeric integral membrane gelatinase belonging to the serine protease family. It is selectively expressed in reactive stromal fibroblasts of epithelial cancers, granulation tissue of healing wounds, and malignant cells of bone and soft tissue sarcomas.
This protein is involved in the control of fibroblast growth and epithelial-mesenchymal interactions during development, tissue repair, and epithelial carcinogenesis.
