Specificity Comments:
FAP (fibroblast activation protein) is a cell surface glycoprotein and serine protease expressed primarily in fetal mesenchymal tissues and epithelial cancer fibroblasts. In cancer, FAP promotes cellular proliferation, while in embryonic development it aids in tissue remodeling. FAP acts as an integral membrane gelatinase composed of N-glycosylated, proteolytically inactive subunits. Its expression on chondrocyte membranes is upregulated by IL-1 and OSM and is increased in osteoarthritic patients, co-localizing with MMP-1, MMP-13, and CD44 variants (v3, v7/8). Mice lacking all copies of the FAP gene are fertile and show no developmental defects or altered cancer susceptibility.
Target Information
The FAP gene encodes a homodimeric integral membrane gelatinase belonging to the serine protease family. It is selectively expressed in reactive stromal fibroblasts of epithelial cancers, granulation tissue of healing wounds, and malignant cells of bone and soft tissue sarcomas. This protein is involved in fibroblast growth regulation and epithelial–mesenchymal interactions during development, tissue repair, and epithelial carcinogenesis.
